Preparation | 5-bromopyrimidine -4-carboxylic acid (prepared according to the method described in U.S. Patent 4,110,450)(1.0 equivalents, 6.14g,30.2mmol) is suspended in CH2Cl2(100ml). Oxalyl chloride (1.1 equivalent, 2.9ml, 33.0mmol) was added, followed by 2 drops of DMF. Stir the mixture overnight at room temperature and remove volatiles in vacuum. The residue was absorbed into MeOH(50ml) and heated. After evaporating MeOH in vacuum to , the compound is dissolved in CH2Cl2 and loaded on a pre-filled silica gel column. Use 20% ethyl acetate in hexane to elute the substance. The solvent was evaporated to obtain methyl 5-bromo-4-pyrimidinic formate, which is a light orange crystalline solid. |
